The paper deals with the use of microsatellite genetic markers for gene diversity assessment of Rupicapra rupicapra. This paper focuses on methods and laboratory protocols.DNAs were extracted from various types of biological material (liver, lungs, heart, tendon, fat, muscle ). The quality of DNA has been tested on agarose gel electrophoresis.The DNA samples were subjected to PCR reactions with specific primers designed and applied for chamois. Microsatellites were electrophoresed on 6% polyacrylamide gels and visualised by silver staining. After silver staining, gels were dried, scanned and visually evaluated. The results of PCR reactions from faeces were not interpretable but still can be used as information for further research.All loci, except one (P17), were polymorphic. Mean value of average number of observed alleles (N.) in six loci was 7,6667 ± 2,3381 and average effective number of alleles (N) was 2,9572 ± l. Mean value of observed heterozygosity (H.) was 0,6289 ± 0,3421 and expected heterozygosity (H) 0,5908 ± 0,2337.For the management of chamois in the High Tatras we suggest: to take measures against reducing population size of chamois, to avoid introducing Alpine chamois, to investigate the reasons for occupying forest habitats by local population of Rupicapra rupicapra tatrica , to consider possibility of Rupicapra rupicapra tatrica introduction to other localities as (e.g. The Low Tatras areas of Kráµova hoµa and Orlová),to consider ex situ conservation in case of dramatic drop in population size of Rupicapra rupicapra tatrica.
