Amperometric acetylthiocholine biosensors were fabricated by entrapping acetylcholinesterase into electropolymerized film of polypyrrole on platinum wire (Pt) and fluorine-tin-oxide (FTO) coated glass electrode. In this study, acetylthiocholine was used as substrate and thiocholine released from the enzymatic hydrolysis was electrochemically oxidized giving anodic current at +600 mV. The enzyme electrodes using a platinum wire as working electrode showed an expanded linear response of 1*10**(-6) to 1.5*10**(-3) M acetylthiocholine and a lower detection limit of 1*10**(-6) M acetylthiocholine, compared with the enzyme electrodes using a FTO-coated glass as working electrode that gave a linear response in the range of 1*10**(-5) to 5*10**(-4) M and a detection limit of 1*10**(-6) M acetylthiocholine.
