Boar taint is the unpleasant odour and flavour to urine-like of the cooked pig meat that is primarily caused by androstenone and skatole deposition in adipose tissue. Although the biochemical processes are well known, their genetic bases are still not clear. We have carried out a differential gene expression analysis between uncastrated pigs of both sexes using Affymetrix microarray and quantitative RT-PCR in order to investigate the expression pattern of genes related with androstenone and skatole removal in hepatic tissue. In total we have found 391 differentially expressed probes between both sexes, 11 related with androstenone and skatole metabolism, four encoding the main androstenone and skatole degradation enzymes. Androstenone degradation is performed in liver by three main enzymes, 3beta-HSD, SULT2A1 and SULT2B1. In our study we have found a lower expression level of 3beta-HSD (0.17X) and SULT2A1 (0.53X) genes in males than in females. In the same way, genes codifying enzymes that participate in the skatole degradation appeared downregulated in males (0.24X for CYP2E1, 0.06X for CYP2A6 and 0.13X forSULT1A1). Moreover, in previous studies it has been suggested that CYB5 enzyme participates in the androstenone synthesis in testicles but it could also participate in steroid removal in liver and it has been proposed as the enzyme connecting androstenone and skatole metabolisms. In the present study we have also found a lower expression level of CYB5 (0.53X) in males that in females. These results may indicate that males have a negative regulation of the expression levels of genes related with the degradation of the main causal components of the boar taint.
