Evalution of ila4 and ifn-r gene expression in vaccinated cattle against theileria annulata and non-vaccinated animals by quantitatvie

The objective of this study was to evaluate the cytokine gene expression (IL-2, IL-4 and IFN- gamma) of vaccinated cattle against bovine theileriosis in comparison with non-vaccinated animals. Evaluation of immune system of vaccinated cattle is too important for studying the immune status of inoculated animals, immunity duration and determination of immune response profile. Cell mediated immunity is the effective response in theileria annulata infection, therefore cytokine study is important. SqRT-PCR was selected as a high sensitive method to detect mRNA transcripts of IL-3, IL-4 and IFN- gamma genes in transcription level by using HPRT gene as internal control gene. 32 animals selected in an intensive dairy cattle Holstein breed farm.They divided in two groupsfirst group includes 22 vaccinated cattle against bovine theileriosis ecently, second group with 10 non-vaccinated heifers as negative control. PBMCs insolated from two groups restimulated with specific theileria annulata antigen as well as positive and negative controls (PHA stimulated and non-stimulated cells respectively) for cytokine gene transcription. The result of sqRT-PCR showed high expression of IL-2 and IL-4 genes in vaccinated animals in response to Theileria antigen, whereas IFN-gamma showed very sligh increasing level of expression, in addition non-vaccinated cattle exhibited no difference in cytokine transcripts in response to Theileria specific antigen. High relative levels of IL-2 and IL-4 gene expression represent Th1 and TH2 profiles in vaccinated cattle in response to Theileria antigen. This in vitro study is the first report of Th1/2 patern in re-stimulated PBMC2 of vaccined immune cattle determined by showing the elevated cytokine ene transcripts in response to Theileria specific antigen. These findings might be important in determining the immune system status of vaccinated cattle against bovine theileriosis.
