Purification Of Toxic Fractions From Odontobuthus Scorpion Venom

Toxic peptides are the main factors in scorpion venom cause toxicity, (their amount being 1% of total venom). In this rese(lfch toxins of Iranian scorpion Odontobuthus Doriac venom have been isolated. At first the crude venom was dialysed against distilled water and eentrifuged in order to separate soluble proteins from the unsoluble mucoproteins. The soluble proteins then were applied on a Sephadex-G50 gel filtration column. The running buffer was 0.1 M Ammonium acetate, PH = 8.6 Toxicity of eluted fractions was examined by i. v injection in mice.The toxic fractions were further analysed by two steps ion-exchange chromatography using DEAE-Sephacel (running buffer being 0.01 M Ammonium acetate, PH = 8.6 with salt gradiant 0.05 M) and Amberlit IRC-50 columns (buffer used was 0.01 M Ammonium acetate, PH = 8.6 with salt gradiant 0.05 M). The purity of the final toxic protein fractions was cheked confirmed by SDS-PAGE. The result of this research shows that purification of the toxic peptides from the crude scorpion venom can be achieved in few steps. Tje purified toxins may then be used for the preparation of antivenom produtes havingless allergenic side effects.Also the toxins may be employed in pharmacological studies of the ion channels on which they affect. Having information abuot the mechanism of action of toxin peptides, it will be possible to design new drugs.
