Kinetics study on the production of microbial coagulant by Rhizopus chinensis Saito 'BIOTECH3273' using batch liquid fermentation of copra meal extract

Growth kinetics of Rhizopus chinensis BIOTECH 3273 using copra meal as substrate was determined using established models. Effect of copra meal concentration on biomass and milk clotting enzyme production, sugar and protein concentrations were also determined. Number of continuous backmix reactors was also calculated. Production using Rotary Shaker Set-up showed that milk clotting enzymes produced using 4%, 6%, 8% and 10% copra meal were higher compared to 4% wheat bran. Enzyme production also increased with increasing copra meal concentrations. Enzymes produced during the first and third day of fermentation did not vary significantly while production peak occurred on the second day 47%, 6% and 8% were used for the 2-L batch production. Statistical analysis of the 2-L process showed that protein and biomass of 4% wheat bran did not vary significantly from 4% copra meal but enzyme and most parameters varied significantly from 4%, 6% and 8% copra meal. The Tessier Kinetic model described the organism's growth obtaining 75.19/hr and 0.0001 mg/ml for µmax and ks, respectively. No inhibition effects were found upon using copra meal. Two reactors were needed for continuous production of 4% wheat bran while only one was needed for all copra meal concentrations.
