Utilization of HPLC-ED techniques for the screening of functional foods from resources of biodiversity | Využití metod HPLC-ED při vyhledávání zdrojů funkčních potravin v rezervách biologické diversity

Within the interdisciplinary cooperation plants belonging to less common fruit species, ex situ collected in germplasm area of Faculty of Horticulture, Mendel University of Agriculture and Forestry in Brno have been evaluated. Partial aims of the work cover the introduction of not commonly used crop with result to laicize health benefits, straightly related to the fruit consumption. Multipurpose potential of the crop from the germplasm have been evaluated in the reason to refer the excessive tendency to supplant the fruit of synthetic food additions. The main goal of the work was focused on the assay development for detection of phenolic antioxidants. According to the results from total antioxidant capacity (TAC) assay, five varieties or genotypes includes the crop from (Crataegus pinnatifida Bunge), (Amelanchier alnifoia Nutt.) Martin, (Amelanchier alnifoia Nutt.) Northline, (Lonicera edulis Turcz. Ex. Freyn) V/8 Sinoglaska and (Lonicera kamtschatica L.) L-K1-21, were evaluated in term of their therapeutic potential. Injected extracts were investigated due to separation of target molecules with using HPLC system coupled with electrochemical (ED) detector and the UV-VIS detector. For relevant expression of present flavonoid content, two of principally different assays were applied. The method results were compared each other. The separation was initiated by isocratic elution of mobile phase; phenolic content was identified from retention of phenolic standards. Significant levels of rutin and quercitrin, both with high level of antioxidant activity, have been observed. The samples showed up high levels of 4- aminobenzoic acids, the highest content of polyphenols was represented by gallic acid. In accordance to results observed, the sequence of HPLC-ED assays was followed by microbiological analysis. In addition, the fruit profiles will be investigated from pharmacological point of view.
