The aim of our study was to compare spore reticulum height from analyzed truffle carpophores collected during summer (August), autumn (October) and winter (December) and to determine application of microscopic spore analysis for distinguishing of Tuber aestivum and T. uncinatum. Our measurements confirmed statistically highly significant differences between spore reticulum height from summer against autumn and winter spores. Statistically insignificant difference was found between spore reticulum height from spores analyzed in fruit bodies collected in autumn and winter. High values of coefficient variation for measured data (v%=22-29%) indicate that spore microscopic analysis is applicable only for larger data files. Individual spore measurements manifested high variability even within the same fruit body. Analysis of samples collected in winter revealed presence of morphologically unripe - colourless spores in asci, what has high importance for optimizing inoculating techniques for production of mycorrhized seedlings and for conservation of truffle genetic resources.
