Modification of Tetrazolium Test methodology for determining viability of pepper seeds | Biber tohumlarında canlılık tespitine yönelik kullanılan Tetrazolium Test metodunun modifikasyonu

Alternative procedure for tetrazolium (2, 3, 5 triphenyl tetrazolium chloride) test to determine viability of pepper (cvs. Kandil Dolma and Yalova Yağlık-28) seeds were studied in this research. In the pretreatment of work, three lots of cvs. Kandil Dolma and Yalova Yağlık-28 were hydratated (3h at 45°C) followed by staining for 2 and 3 h in a 1% TZ solution at 35, 40 ve 45°C. Results of viability were associated with germination and seedling emergence test.

Araştırmamızda, ülkemizde de yoğun olarak yetiştiriciliği yapılan Kandil Dolma ve Yalova Yağlık-28 çeşidi biber tohumlarında, canlılık tespitinde kullanılan tetrazolium (2, 3, 5 triphenyl tetrazolium chloride) (TZ) testi için en uygun prosedür belirlenmeye çalışılmıştır. Denemede biber tohumları (her çesit için 3 er parti) 45°C'de 3 saat nemli ortamda tutulduktan sonra 35, 40 ve 45°C de %1'lik TZ solüsyonunda 2 ve 3 saat bekletilmiştir. Elde edilen sonuçlar, korelasyon analizi yapılarak çimlenme ve fide çıkış oranları ile ilişkilendirilmiştir.
