Chino. 采用PCR-SSCP技术对荷斯坦牛和娟-荷F1牛2个群体奶牛的热应激基因(HSPA8和HSPA3)进行了单核苷酸多态性(SNP)分析,对部分基因型个体的PCR产物进行测序和序列比较,并分析了标记基因型与奶牛血清生化指标间的相关性.测序发现:HSPA8座位存在T缺失突变以及C→G颠换和T→C转换突变;HSPA3座位发现C→A颠换突变.相关性分析表明,HSPA3和HSPA8位点多态性与2个群体奶牛血清中总抗氧化能力(T-AOC)没有关联,而与血清中的总超氧化物歧化酶(T-SOD)活力、丙二醛(MDA)含量有一定的相关.[著者文摘]
Mostrar más [+] Menos [-]Inglés. The heat tolerant traits were studied in Holstein and Jersey x Holstein F1 dairy cows. Single nucleotide polymorphism (SNP) of heat stress protein gene (HSP) was analyzed by PCR-SSCP and gene and genotype frequencies were calculated for the polymorphic loci in HSP gene. Effects of genotypes in different locus on heat tolerant traits were estimated. The results indicated that T deletion mutations (C→G, T→C) in HSPA8 and (C→A) in HSPA3-exonl were detected. Correlation analysis showed that there was no relationship between the polymorphisms of HSPA3 and HSPA8 and total antioxide cables (T-AOC) ; However there were pos- itive relationships between polymorphisms of HSPA3 and HSPA8 and malondialdehyde (MDA) content and total superoxide dismutase (T-SOD) activity.
