Effects of SNP in the exon 3 of Ghrelin gene on carcass traits in Chaohu duck | 巢湖鸭Ghrelin基因外显子3的单核苷酸多态性及其对屠体性状的影响_

[目的]检测巢湖鸭Ghrelin基因外显子3的单核苷酸多态性(SNPs),并分析其与巢湖鸭屠体性状的关系。[方法]以巢湖鸭为试验群体,采用PCR_SSCP和DNA测序的方法检测鸭生长素(Ghrelin)基因外显子的3SNPs,并将所发现的SNPs与胴体重、半净膛重和全净膛重等屠体性状进行相关性分析。[结果]巢湖鸭外显子3 第54 bp处发生G→A的沉默突变,检测到AA、BB和AB 3种基因型,该突变位点对巢湖鸭胴体重影响极显著_(P0.01),对半净膛重和全净膛重有显著影响_(P0.05),BB基因型在胴体重、半净膛重和全净膛重上显著高于AA型和AB型(P0.05)。[结论] Ghrelin基因多态性与巢湖鸭的部分屠体性状存在显著相关性,对B等位基因进行选择将有利于提高巢湖鸭的屠体指标。_

[Objective]To analyze the polymorphism of Ghrelin gene of Chaohu duck and the relationship with its carcass traits.[Method] PCR-SSCP and DNA sequencing technologies were applied to detect single nucleotide polymorphism (SNP) of exon 3 of the Ghrelin gene in Chaohu duck and the relationship between the polymorphisms of Ghrelin gene and carcass traits (including carcass weight, half-eviscerated weight and carcass net weight, and so on) of Chaohu duck was investigated.[Result] The results showed that, one silent mutation G→A was found at 54 th bp in the third exon of Ghrelin gene and three different genotypes were identified as AA, AB and BB. This base mutation showed different effects on the carcass traits, the extreme significant effect of various genotypes was yielded on carcass weight (P0.01), and differences were significant on half-eviscerated weight and carcass net weight (P0.05) of different genotypes. The values of BB genotype were significantly higher than that of AA and AB genotypes (P0.05) according to above traits.[Conclusion]There was significant correlation between the genetic polymorphisms of Ghrelin gene and the carcass traits and the allele B was valuable for increasing some carcass traits._
