Chino. 利用双抗夹心酶联免疫吸附测定(DAS-ELISA)的方法对甘肃出入境南瓜、西葫芦种子及采自河西地区显症病株叶片进行检测,在种子及病叶组织中均检测到ZYMV病毒,其中南瓜种子带毒批次占12.5%,西葫芦种子带毒批次占11.8%。根据已报道的小西葫芦黄花叶病毒(Zucchini yellow mosaicvirus)基N组核苷酸序列,设计引物扩增其外壳蛋白(CP)基因,以ELISA阳性种子或病叶组织总RNA为模板,进行RT-PCR扩增,对预期大小的扩增产物进行测序,结果表明扩增获得的核苷酸序列与世界各地的ZYMV分离物CP基N具有高度一致性,综合ELISA检测和RTPCR的结果,确定南瓜、西葫芦种子可携带ZYMV,且ZYMV是侵染甘肃瓜类作物的重要病毒种类。[著者文摘]
Mostrar más [+] Menos [-]Inglés. Zucchini yellow mosaic virus (ZYMV) is an economically important virus of cucurbit crops. Using DAS- ELISA and RT-PCR, ZYMV was detected from the leaves of Cucurbita moschata and C. pepo showing symptoms or from seed samples. A total of 58 seed lots were tested. The infected rates of ZYMV were 12.5 % in squash seed lots, and 11.8% in zucchini seed lots. A specific fragment of about 280 bp in length of the coat protein (CP) gene was obtained by RT-PCR and sequenced. Sequence analysis showed that it shared 93%--98% nucleotide acid identities with the published sequences. These results indicated that ZYMV is one of the most important viruses threatening the cucurbit crops (e. g., squash, watermelon, melon and cucumber) in Gansu Province.[著者文摘]
