Amino acid polymorphisms in the ovine priori protein gene at codons 136,154, and 171 affect the susceptibility of sheep to scrapie, a naturally occuring form of transmissible spongioform encephalopathy (TSE). The aim of this work was to evaluate the frequencies of PrP polymorphisms at codons 136 (A/V), 154 (R/H), and 171 (Q/R/H) in the subpopulation of autochthonous sheep of Valachian breed, which belongs to endagered sheep breeds in Slovakia. Genotyping was performed by the PCR-PIRA method. The most frequent haplotype was ARR (65.7%), the frequency of other haplotypes was much lower; ARQ (22.9%), AHQ (0%), ARH (10%), VRQ (1.4%). Accordingly, also the genotype ARR/ARR (51.7%) was the most frequent, while the genotypes ARR/ARQ (25.7%), ARQ/ARQ (8.57%), ARH/ARH (8.57%), ARQ/ARH (2.86%), and ARR/VRQ (2.86%) showed lower frequency. All genotyped rams were the ARR/ARR genotype. No other possible haplotypes and genotypes were detected in the analysed animals.
