Isolation, identification and partial nucleotide sequence of a flexous rod virus causing yellow spot mosaic of purple passionfruit

Passiflora edulis in field of royal project foundation revealed problem, woodiness disease which reduced product and quality of fruit. This project was study for identification virus cause disease. Test host range found chlorotic local lesion on Chenopodium quinoa and C. amaranticolor. Inoculation on leaves Leguminosae showed local lesion on inoculated leaves and yellow vein symptoms on tip leaves. Physalis angulata showed yellow, distortion, malformed leaves and yellow mosaic on N. benthamiana. Particle virus were flexous rods, size of 650 nm. long and found inclusion bodies protein in cytoplasm of infected cell. Cloning coat protein gene isolate LG2 and LG3, which isolated from grafting on healthy passionfruit, had 819 nucleotides, translate into protein 272 residues. Phylogenetic tree was similarity CP amino acid sequences with TeMV isolate from veitnam showed 87 percent homology and were grouped with PWV-isolate from Australia. This results showing virus cause woodiness disease was TeMV both isolate LG2 andLG3.
