In the present study, callus was induced from explants of in vitro raised plants obtained from self pollinated seeds of mulberry variety Berhampore local when cultured on MSBM supplemented with 2,4-D (2 mg/l), CM (10 /) and CH (1 mg/l). After three weeks of subculturing on the same medium the callus proliferated further into profuse compact callus with the development of tiny green primordia. This morphogenic callus developed 5-6 multiple shoots within six weeks when transferred on MSBM supplemented with BAP (2.5 mg/l), CM (10/) and CH (1 mg/l). In the present study it was found that callus can be maintained up to 6 months on MSBM with 2,4-D (2 mg/l) and CM (10 /). For the first time, repeatable protocol was developed for differentiation of shoots from the stem callus for this variety. The differentiated shoots were transferred to rooting media and the best rooting was observed on MSBM supplemented with NAA (1 mg/l). The rooted in vitro plants were successfully hardened and transferred to the field.
