The present study is concerned mainly with some effects of Medroxyprogesterone acetate (DMPA) on chromosome of the rat bone marrow cells. The rats injected with DMPA in different dosages : 0.0285, 0.0571, 0.1142 and 0.1428 mg/gBw, then left for 6, 12, 24 and 48 hours followed by harvesting the bone marrow cells. The specimens were examined and photographed with a compound microscope. It was observed that DMPA could induce the majority chromatid breakage, dicentric, polycentric, acentric and ring chromosome. When the dosages of DMPA were increased, the number of these were increased too. However the periods from injecting DMPA into the rat to harvesting the bone marrow cells were increased, the number of chromosomes were increased, but acentric chromosomes were decreased. The dosages of DMPA and periods from injecting DMPA into the rat to harvesting the bone marrow cells would induce the damage and repair of DNA on chromosome as well. Based on these evidences DMPA is both a mutagenic and carcinogenic agent.
