Mycobacterium sp. were isolated from Siamese fighting fish (Betta splendens Regan) that showed or did not show external signs in infected fish which include exophthalmia, scale defects, loss of scales, fin rot and ascites. The isolates were acid fastness, rod shape, grew on Ogawa egg media within 3-5 days, photochromogenic and produced mycolic acid. DNA was isolated by modify method of Ezaki. Cell wall of mycobacteria was disrupted by ultrasonification and lysed by lysozyme. DNA was extracted by phenol and chloroform extraction, ribonuclease digestion and ethanol precipitation. Mole percent guanine plus cytosine was calculated from the thermal melting point of DNA in SSC buffer. The average GC content determined was 63.66 percent with standard deviation of 1.2.
