An effective, quick and simple method of differentiation of pebrine spore from non pebrine spore and artefacts during mother moth examination, was a prerequisite when a large number of silk moths were examined for pebrine spore in grainage centres. In the field condition, examination, for pebrine spore in homogenate of mother moth without centrifugation, filtering and staining posed problem of precise detection of pebrine spore due to the presence of such artefacts and non pebrine spores. A simple positive staining method was developed and described, which differentiated pebrine spore Nosema bombycis from non pebrine spores in silkworm Bombyx mori L. The method increased the visibility of the pebrine spores from spore resembling non pebrine artefacts by differential staining. One percent crystal violet stain with a pretreatment with ten percent potassium permanganate solution (KMnO4) stained the spores in less than two minutes. The paper describes the methodology of staining, results and its advantages.
