CSN3 gene was suggested as a candidate gene for milk yield, percentage of protein and fat content. CSN3 variant B is reported to be favorable for milk quality and is considered to be included in breeding strategies of dairy animals. The aim of this paper was to use the PCR - RFLP as a fast and efficient method to detect the genetic variants of CSN3 gene in dairy cattle followed by use of restriction enzymes HindIII for genotyping. The polymorphism of CSN3 gene was studied in a group of 80 cows (crossed Slovak spotted x Holstein of different share). Genotype AA was the most frequent one with observed frequency of 57.5%, while the frequency of the AB genotype was lower (36.25%). The BB genotype was represented with the frequency of 6.25%. The frequency of alleles A was 0.7563 and B was 0.2437.
