Effect factors of porcine oocytes in vitro maturation | 猪卵母细胞体外成熟的影响因素

Chino. 摘 要:通过采集屠宰场废弃猪卵巢,体外分离和培养猪卵母细胞,探讨基础培养基、激素、卵泡直径大小及卵丘细胞层数对猪卵母细胞体外成熟的影响。结果表明:改良TCM199(mTCM199)较NCSU-23培养猪卵母细胞成熟率显著提高(P〈0.05);基础培养基中添加PMSG、hcG和pFF,卵母细胞体外成熟率(80.63%)显著高于仅添加其中一种;从卵泡直径3~6mm和大于6mm卵泡获得的卵丘卵母细胞复合体(COCs),其体外培养成熟率(83.33%和76.43%)极显著高于卵泡直径在3mm以下获得的COCs成熟率(30.94%);根据卵丘细胞层数把COCs分为A、B、C3级,其中A级和B级卵母细胞成熟率差异不显著,但都极显著高于C级卵母细胞的成熟率。
Mostrar más [+] Menos [-]Inglés. In present study, porcine ovaries were obtained and aspirated. Cumulus-oocyte complexes were followed by in vitro culture until the presence of the first polar body(pb I ). We investigated the effects of basic culture medium, hormone, follicle diameter, and oocytes wrapped cumulus ceils on in vitro maturation of pig oocytes for establishing good in vitro culture system. The results showed that the mature rate of oocytes with mTCM199 was significantly higher than NCSU-23(P〈0. 05). Based on the results of experiment 1, the maturation rate of cumulus-oocyte complexes in maturation media with PMSG,hCG and pFF were 55. 90%, 58. 16%, and 51. 50%, respectively, showing that the combined treatment group (80. 63%) was significantly higher than those of single treatment groups. There were no significant differences in the maturation rates of the ovary follicles larger than 6 mm (76. 43%) and 36 ram(83.33%) in diameter, but both were significantly higher than the ovary follicles less than 3 mm in diameter(30. 94%, P〈0. 01). The cumulus-oocyte complexes were divided into level A, B and C according to layer of cumulus cells, which were cultured in vitro matured for 44 h,the maturation rates of level A and B were not significant (P〉0. 05), but the both were significantly higher than oocytes of level C(P〈0. 01).
