Effects of HSP70 gene polymorphism on thermo tolerance traits of yellow feather dwarf broiler | HSP70基因多态性对矮小型黄羽肉鸡热耐受性的影响

Chino. 采用PCR产物直接测序和PCR-RFLP检测的方法,选取HSP70基因的C.-69AG、C.+258AG、C.+276CG、C.+909CA、C.+1044AG、C.+1437CT和C.+1476CT 7个多态位点,分别在15和35℃条件下对矮小型黄羽肉鸡纯系母鸡进行与热耐受性相关的部分生理与免疫指标的关联分析。结果表明:热应激状态下(35℃),C.+276CG位点与CD3+T细胞含量显著相关(P0.05),GG基因型个体的CD3+T细胞含量极显著高于CC基因型个体(P0.01)、显著高于CG基因型个体(P0.05);C.+1437CT位点与CD4+T细胞含量极显著相关(P0.01),CC基因型个体CD4+T细胞含量极显著低于CT和TT基因型个体(P0.01)。C.+1476CT位点的CT基因型个体H/L值显著低于CC基因型个体(P0.05)。常温状态下(15℃),C.-69AG位点的GG基因型个体CD4+T细胞含量显著高于AG基因型个体(P0.05);C.+1044AG位点与CD4+T细胞含量显著相关(P0.05),AG基因型个体CD4+T细胞含量显著高于AA基因型个体(P0.05);C.+1437CT位点与CD3+T细胞含量、皮质酮值显著相关(P0.05),CC基因型个体的CD3+T细胞含量和皮质酮值都显著高于CT和TT基因型个体(P0.05)。与常温状态相比,各位点同一基因型在热应激状态下T3水平、H/L值和CD4+T细胞含量都显著升高,皮质酮含量和CD3+T细胞含量都显著下降。C.-69AG、C.+276CG、C.+1044AG、C.+1437CT和C.+1476CT 5个位点与部分生理、免疫指标显著相关。其中C.+1437CT位点与多个生理、免疫指标都有显著或极显著相关,表明该位点对矮小型黄羽肉鸡的热耐受性具有潜在的遗传效应。

Single nucleotide polymorphism(SNP)C.-69AG,C.+258AG,C.+276CG,C.+909CA,C.+1044AG,C.+1437CT and C.+1476CT of HSP70 gene of yellow feather dwarf broiler have been identified by direct sequencing and PCR-RFLP so as to further perform their associations with part of physiological and immune indices.The results indicated that,under heat stress(35℃),SNP C.+276CG was significantly associated with CD3+T cell levels(P0.05),and the CD3+T cell level of GG genotype was significantly higher than CC(P0.01)and GG genotypes(P0.05).SNP C.+1437CT was significantly associated with CD4+T cell levels(P0.01),with a lower CD4+T cell level for CC than for CT and TT genotypes(P0.01).The H/L value for CT of SNP C.+1476CT was lower than CC genotype(P0.05).In case of ordinary condition(15℃),the CD4+ cell level for GG of SNP C.-69AG was higher than AG genotype(P0.05).A mutation at C.+1044AG was significantly associated with CD4+T cell levels(P0.05),with a higher CD4+T cell levels for the AG than for the AA genotype(P0.05).SNP C.+1437CT was significantly associated with CD3+T cell levels and cortisone(P0.05),with higher values for CC than other two genotypes.Compared with the ordinary condition(15℃),the T3,H/L value and CD4+ for the same genotype in SNP were significantly increased under heat stress condition,and the contents of corticosterone and CD3+ were decreased significantly.SNP C.-69AG,C.+276CG,C.+1044AG,C.+1437CT and C.+1476CT of HSP70 gene were significantly associated with part of physiological and immune indices.Moreover,SNP C.+1437CT has significant associations with some physiological and immune indices,which indicates its potential genetic effect on the yellow feather dwarf broiler thermo tolerance.
