Chino. 针对马铃薯块茎含有多糖多酚的特点,选用3种快捷的商品化提取试剂提取马铃薯块茎总RNA。结果表明:3种商品化试剂均可不同程度的提取马铃薯块茎总RNA,提取的总RNA的28S、18S和5SrRNA条带清晰可见,但纯度和浓度存在较大差异;其中用Trizol裂解加柱吸附法提取的马铃薯块茎总RNA纯度较高,但浓度较低,经SMART PCR和LD-PCR检测可扩增出马铃薯块茎腺苷葡萄糖磷酸化酶小亚基(sAGP)基因和全长双链cDNA,可用于后续分子生物学试验。
Mostrar más [+] Menos [-]Inglés. Three commercialized reagents were taken to isolate total RNA from potato tuber for its higher concentrations of polyphenolcs and polysaccharides. The results showed that total RNA could be extracted from potato tuber in some degree using the three commercialized reagents, and the 28S, 18S and 5S rRNA bands were clearly visible, however, RNA purity and concentration were quietly different. The extracted RNA with Trizol lysis solution and absorption column had higher purity and lower yield. Full-length ds-cDNA and sAGP gene of potato tuber were detected by SMART PCR and LD-PCR, which could be used for downstream molecular experiments.
