Evaluation of interaction of corn hybrids and their inbred lines to corn common smut (U.maydis)

For resistance evaluation of common corn hybrids and related lines against common smut, the fungous teliospores were collected from naturally infected ears of corn fields. Teliospores were surface- disinfested in copper sulfate 1.5% for 6 hour on shaker, then washed three times with distilled water and spread on Potato -dextrose agar (PDA) containing 106 gil of dextrose in petridishe, and incubated for 5 days at 23°C, sporidia were collected by distilled water containing 2 drops of Tween 20 per 100ml and gently scraping the agar media surface, the suspension was filtered through a four layers of chease cloth , sporidia were counted in a hema cytometer and was adjused to 106 sporidia per millititer. Corn hybrids such as : 704, 711, 604, 301 , 108 and inbred lines N017, K722, 861, B73 , LI05 and TVA were grown in 2: sand l:soil mixture in 30-cm. diameter (3 plants per pot) in glass house, the plants were inoculated by using a syringe to inject Zml of the sporidia in the leaf whorl at 4-6 leaves stage. The first symptoms of the disease was observed as sunken spots in leaves, but gall rarely developed in leaves becaus of the plants couldn'g grow enough in green house, so we sugested field trial for evaluating reaction of corn hybrid to the diseas.
