Micropropagation hybrid of (Populus euphratica× P. alba)by tissue culture

In order to micropropagate the hybrids Populus euphratica×P. alba andP. alba×P. euphratica, shoot tips were harvested from research nercery of Biotechnology Department, Research Institute of Forest and Rangeland. The buds were established after sterilizing by 0.1%W/V mercuric chloride for 1 minutes on the MS medium containing 0.5mg/l BA and 0.01mg/l IBA. After that, the shoots of both hybrids were transferred separately in a completely randomised design with 9 treatments on three different media (MS, WPM and DKW) and with different ranges of hormones (IBA, 2iP and BA) in 3 repetition and 3 periods (every period equal a month and each repetition include 30 explants). Data were analyzed by mean of mini-tab software at %5 confidential level. The best treatment with the highest level in coefficient shooting and the best growth in length were obtained from DKW medium which compleated with BA and 2iP by concentration of 0.5mg/l and 0.01mg/l IBA. Rooting of shoots occurred in DKW medium, without hormone after two weeks. These plantlets were established in polyethylene caps jam buttel, containing a light soil (peat, homus and sand in equal percentages) and kept in a germinator in order to acclimatization. Finally, they acclimatized successfully and transferred to field after 40 days.
