Annual technical report 2012 January-December [UPLB-BIOTECH National Immunology Lab.]
2012
Tavanlar, M.A.T.
The National Immunology Laboratory continued the production of antigens from Abaca Bract Mosaic Virus (ABMV), Banana Bunchy Top Virus (BBTV) and Banana Bract Mosaic Virus (BBMV). The purification procedure for the viruses had to be modified because of the unavailability of an ultracentrifuge. Results from successive filtration of sap from BBMV-infected banana showed flexuous rods typical of BBMV. Due to the difficulty in procuring aflatoxin B1-bovine serum albumin conjugates that are needed as antigens, the isolation of aflatoxin from toxigenic fungi was performed in order to augment the toxin stock available for testing of the generated antibodies. The production of immunogens from haptens of chlorpyrifos was continued for the externally-funded project on pesticide residue analysis. Polyclonal antibodies were produced from the above-mentioned antigens and were characterized by indirect ELISA for titer, purified and conjugated to enzymes to serve as tracers in the development of protocols for direct microplate and membrane strip immunoassays. Monoclonal antibody production was deferred pending the result from the polyclonal antibodies. The myeloma cell line NS1 was routinely transferred to fresh media with supplements to invigorate the cells. A total of 594 samples of abaca and banana were submitted by clients for viral indexing. Sixty-seven samples of corn, rice, peanut and peanut butter from a client from the University of Northern Philippines were analyzed for aflatoxins using thin layer chromatography. Red tide monitoring was done in the coastal areas of Bacoor, Las Piñas, Parañaque, Sangley Point and in five towns in Bataan. Although mussels and oysters from the Bataan sampling sites were negative in the mouse bioassay test, sample of green mussel or 'sulib' showed lethality corresponding to 0.56 to 0.98 mouse units. Furthermore, the microbiological load of the seawater and shellfish samples was very high. The gas chromatograph and greenhouse no. 2 as well as two vehicles were repaired during the year. The gross receipts from services rendered to clients and sale of laboratory mice amounted to forty-nine thousand and eighty pesos (PhP49,080.00).
