Isolation of bioactive terpenoids from leaves of the olive tree using preparative liquid chromatography

The aim of this study was a contribution to the preparative isolation of known triterpenoids from the leaves of olive trees, using fast, inexpensive and environmentally friendly methods of extraction and identification of the targeted compounds. The extraction was performed by various solvent systems and conditions and the extracts were then purified and analyzed. The research focused on the extraction of triterpenic acids from the olive leaves using a fast, inexpensive and environmentally friendly method. The triterpenic acids mainly involved in the project are oleanolic, ursolic and maslinic acid (expressed as UA, being the standard available). The extracts are ethanolic, alkaline (x 2), ethanol alkaline and trizma. Some of them were fractionated on column chromatography and analyzed in GC-MS and HPLC-MS-ESI+. The results showed that these triterpenic acids are found in the olive leaf extracts obtained throughout the study. Although the separation and identification of these acids is considered challenging due to the similarity in their structure, three of these acids were detectable and identified in olive leaves. The extractions were to a large extent adequate with the aim of the study regarding the speed of the extraction, its cost and its non-polluting role.
