Study on extraction process of free amino acids from Smilacina japonica | 鹿药游离氨基酸提取工艺

Chino. 以鹿药为试验材料,采用超声波辅助浸提法,通过单因素试验和正交试验,分别对料液比、提取时间、提取温度及超声功率等因素进行考察,优选鹿药游离氨基酸的最佳提取工艺条件。同时,利用氨基酸自动分析仪对鹿药游离氨基酸进行组分及含量测定分析。结果表明:料液比1:30、提取时间30min、提取温度40℃、超声功率80w,超声提取鹿药游离氨基酸的提取率最高。鹿药由16种不同氨基酸组成,其中必需氨基酸占总氨基酸含量的47.93%。料液比对游离氨基酸提取率影响较显著,且游离氨基酸含量随鹿药采收期的延长呈下降趋势。

To optimize the extraction technology for free amino acids in Smilacina japonica, taking Smilacina japonica as raw materials, the effects of material-liquid ratio, extraction time, extraction temperature and ultrasonic power on extraction rate of free amino acids from Smilacina japonica were studied by single factor experiment and orthogonal test. At the same time, composition and content of free amino acids in Smilacina japonica were determined and analyzed by automatic amino acid analyzer. The results showed that the optimum extraction conditions were as follows: material-liquid ratio 1:30, extraction time 30 rain, extraction temperature 40℃ and ultrasonic power 80 W, and the extraction ratio of free amino acids with ultrasonic extraction was the highest. Smilacina japonica is composed of 16 kinds of amino acids, among which the essential amino acids account for 47.93% of total amino acids. The results also indicated that material-liquid ratio had a significant effect on the extraction rate of free amino acids and the content of free amino acids decreased with the extension of harvesting time. These primary research results provide credible foundation for utilization of Smilacina japonica.
