A study on in vitro rapid propagation technology for crisp plum in Guizhou province | 贵州酥李离体快繁技术研究

Chino. 以酥李带芽茎段为外植体进行组织培养,研究了植物生长调节剂组合对腋芽诱导、增殖及生根的影响,初步建立酥李离体快繁技术体系。结果表明,外植体消毒采用1g/LHgCl2浸泡8min的效果较好,适宜腋芽诱导培养基为WPM+1.0mg/L 6-BA+0.5mg/L KT+0.1mg/L NAA,适宜芽增殖培养基为WPM+1.0mg/LKT+0.1mg/L IBA+1.0mg/L GA3,适宜生根培养基为1/2MS+0.4mg/L IBA+15g/L蔗糖,其生根率达95%,移栽成活率在90%以上。

Using the axillary growth regulators on differentiation rapid propagation techniques were buds as explants for tissue culture, the proliferation and rooting of shoots in Crisp established. The results showed that the effects of combinations Plum were investigated of plant, and the optimal disinfection time of explants in 0.1% HgCl2 was 8 min. The optimal medium for initial culture was WPM+1.0 mg/L 6-BA+0.5 mg/LKT+0.1 mg/L NAA. The optimal multiplication medium was WPM+1.0 mg/LKT+0.1mg/L IBA+1.0 mg/L GA3. The optimal medium for rooting was 1/2 MS+0.4 mg/L IBA+15g/L sugar, and its rate for rooting reached 95% and rate of transplanting survival reached 90%.
