Survey of potato fields for identification and dissemination of Globodera sp. in the country

Potato (Solanum tubrosum L.) is one of the important crops which is hosting of a large number of plant pathogens. Among the potato pathogens, Potato cyst nematode (PCN) is one of the important pathogens worldwide. PCN can reduce the yield quantity to less than initial planting seed amount. This pathogen was, for long time, in the Iranian external quarantine list, but unfortunately it could pass the borders along with seed potatoes and was observed in Hamedan province in 2008 for the first time. The first step for disease management is to identify the distribution of the pathogen. For this purpose, a national project was carried out for detection of the distribution of the disease and drawing of the map of infected area in Iran. During 2009-2012, different pototo fields of the Zanjan province was surveyed and 168 samples from soil and potato root systems were collected, transferred to the laboratory and were subjected for isolation of the nematodes using sieves, centrifuge and Fenvik apparatus. After isolation, the nematodes were fixed and used for morphological studies. For further confirmation, about 110 samples of cyst nematodes were transferred to the department of plant nemathology in Iranian plant protection institute. The results showed than none of the isolate cyst nematodes from Zanjan province belonged to the Globodera species. The Isolated cyst nematodes belonged to Heterodera latipons and H. avenae. The high number of cereal cyst nematodes in Zanjan, may be justified by rotation of potato with cereals in this region.
