The study of citrus greening distribution in south of Kerman Province.

HLB (Huanglongbing ex greening) caused by three species of Candidatus Liberibacter asiaticus, Ca. L. africanus and Ca. L. americanus, is the most important citrus disease of the world. The disease is transmitted by two psyllids Diaphorina citri and Trioza erytreae and infected bud woods. One thousand samples were analyzed for HLB.DNA was extracted from the leaves by CTAB methods with minor modifications. DNA amplification was conducted by Conventional PCR using A2 and J5, OI1, OI2c specific primers of the Ca. L. asiaticus. 703 bp and 1160bp bands were obtained from ten samples of the symptomatic tress. 1000 samples from south of Kerman analyzed that 5.1 percent were positive to HLB diseases. The PCR amplicons of 12 Iranian strains with OI1 and OI2c primers purified and sequenced. The Consensus sequences of Iranian strains compared with a Brazilian isolate of Ca. L. asiaticus. They showed 100% homology with standard HLB Asiatic type. However, they had different in symptom between Iranian strains, but not be any different in consensus sequences of ribosomal DNA (OI1 and OI2c). This is the first report of HLB disease in this region and comparison Iranian strains in molecular based levels with Asian type strain of Ca. Liberibacter asiaticus from Brazil.
