Anti-microbial proteins appear the components of protective systems of all pluricellular organisms. Particular place among anti-microbial proteins appertains to plant defensins, possessing fungicidal, insecticidal and antibacterial activity. Thionins appear one of the most important proteins with low molecular weight and rich in sulfur-containing amino acids. Thionins are high toxic to bacteria, fungi and yeasts. Development of the method of determination of thionins' content in aboveground portion of a plant on the example of barley (Hordeum vulgare) seedlings is suggested. The method of extraction of thionins from barley seedlings is presented. It is shown that additional procedures such as heating and disulfide bond reduction as well as sulfhydryl group alkylation are necessary to improve thionins' separation and identification. Antipathogenic properties of thionins extrracted from etiolated barley seedlings are shown with use of Fusarium oxysporum.
