Poultry industry is one the most important industries in Iran on which a lot of money is capitalized. Despite the efforts to develop the gains on production, diseases occur yet. Mycoplasma species are problematic, ever-present agents among which M.gallisepticum and M.synoviae have the ability to damage chicken respiratory organs causing infections, production decline, mortality and condemnation. Great medical expenses are also imposed to the economy due to prevention and elimination. Even though, it is obvious that recognizing the infected broiler breeder flocks and line flocks, and removing them is pretty cheaper and safer. This study is one of the national investigative projects called "Isolation and identification of pathogenic Mycoplasma spices from chicken farms in different country provinces" which was done in collaboration with Veterinary organization. According to a pre-arranged plan, 66 active sites in Tehran, Karaj, Shahriar, islamshahr , varamin , Savojbolagh and Ray were visited samples from 1/746/383 breeder chicken from 465 farms were collected . Using the statistics method 5949 blood samples and pharyngeal swab samples were gathered. Rapid serum agglutination test (RSA) was used on blood samples and swab samples were cultured in PPLO broth and PPLO agar mediums. Isolation and identification of pathogenic Mycoplasams was dong and visiting the fired egg like colonies growing mediums. M.g positive result in blood samples was 6.56 while this number from M.s was recorded 61.82 using the RSA. From 465 farms, after culturing the pharyngeal swabs, it was presented that 137 of them (29.5%) were infected by Mycoplasma. After isolation test and differentiation procedures it was declared that 19 farms (4%) were infected by Mg, 75 farms (16.3%) by Ms and 9 farms (1.9%) were struggling both. Samples from 34 farms (&.3%) were not identified with M.g and M.s anti-serum. According to the results, the breeder farms in Tehran province involvements with Mycoplessmas is strongly confirmed .It is highly recommended to use rapid tests like RSA as screening test and to consider the Molecular methods in isolation before removing the flocks. Key words : Mycoplasma gallisepticum, Mycoplasma synoviae, broiler breeder farms, Rapid serum agglutination test ( RSA), Isolation, Identification.
