Isolation and identification of strains of Mycoplasmas causing contagious Agalactia in sheep & goats, in fars province by using of culture method and PCR.

Isolation and Identification of Mycoplasma agalactiae, from sheep and goats by culture and PCR methods in Fars province. Contagious agalactiae in the form of Mastitis, keratocunjuanctivitis and Arthritis is a disease of sheep and goats, which is caused by Mycoplasma agalactia a gram negative bacterium without cell wall that induces heavy economic losses. The aim of this study was to isolate and identify M.agalactiae from sheep and goats at Fars procince. Clinical samples such as milk, eye swabs and synovial fluids were colected from infected animals. The collected samples were transported to to the laboratory of Razi Vaccine and Serum Research Institute, Shiraz branch, in a medium that contains (PPLO brath, hors serum and yeast extract), at cold conditions. The samples were examind by PCR and culturing on PPLO agar. The results showed that from 98 milk samples 18.36% were positive by PCR and 12% by culture for mycoplasma. Out of 80 eye swabs, 11.25% was positive by culture and the same results were obtained by PCR too. Of synovial fluid, 3 of 8 samples were positive for mycoplasma by PCR, while none of them was positive in culutre. In conclusion mycoplasmosis due to M.agalactia infection is present in the flocks of Fars province. For a quick detedction and epidemiological study of mycoplasma disease PCR was found to be a suitable test than a culture. Because, in cuture, mycoplasma would take time to grow well, while by PCR it could be identify within a few hours. Key wourds: Mycoplasma, agalactia, sheep, goat, culture, PCR, Fars province.
