Study of grain Cd uptake of newly developed high grain Zn breeding lines in rice

Researchers are developing new rice breeding lines with higher grain Zinc (Zn) content. Methods such as agronomic management and plant breeding are used to improve the grain Zn content in rice. Researchers are also concerned about the possibilities of higher grain Cd uptake of the newly-developed Zn-biofortified rice lines. This study examined the effects of genotypes and available soil Cd to the grain Zn concentration and grain Cd concentration of rice. A greenhouse experiment was conducted. Three levels of Cd contamination in soil were used; soils were collected from IRRI, San Leonardo Nueva Ecija (NE) and Sta. Rosa NE. Five genotypes were used in these experiment; Wild Type (WT) - control, IR64-IRS495-234(0), IR64-IRS495-234(1) (transgenic), IR64-IRS495-274(0), and IR64-IRS495-274(1) (transgenic). Agronomic data were collected during the harvest stage. Grain samples were dehulled, polished and analyzed for grain Cd and Zn concentrations. The extractable Cd of IRRI, San Leonardo and Sta. Rosa are 0.19 ppm, 0.22 ppm, and 0.31 ppm respectively. Data showed that transgenic lines significantly increased the grain Zn concentration. The average grain Zn concentration of IR64-IRS495-234(0) in the 3 soils is 27.7 ppm Zn while the transgenic line IR64-IRS495-234(1) had 44.7 ppm Zn. The average grain Zn concentration of IR64-IRS495-274(0) in the 3 soils is 34.4 ppm Zn while the transgenic line IR64-IRS495-274(1) had 53.8 ppm Zn. On the other hand, transgenic lines did not significantly increase the grain Cd concentration. The average grain Cd concentration of IR64-IRS495-234(0) in the 3 soils is 56.8 ppb while the transgenic line IR64-IRS495-234(1) had 56.9 ppb. The average grain Cd concentration of IR64-IRS495-274(0) in the 3 soils is 110.9 ppb while the transgenic line IR64-IRS495-274(1) had 103.0 ppb. The grain Cd concentrations of all breeding lines were below the threshold concentration of 400 ppb.
