Effect of Aronia Extract on the Lymphatic Absorption of Cholesterol and Fat in Rats

Aronia contains various phenolic antioxidants including anthocyanins. Several studies have shown that dietary intake of aronia may prevent hypercholesterolemia. The objective of this study was to investigate the effect of polyphenol-rich aronia extract (AE) on the intestinal absorption of cholesterol (CH) and fat in rats. First, the antioxidant activities of AE were analyzed. Total phenolics amount was 169.79 mg gallic acid equivalent/L and 1,1-diphenyl-2-picrylhydrazyl radical scavenging activity was 23.7%. Next, male Sprague-Dawley rats that were raised individually were cannulated with a lymph cannula in the mesenteric lymph duct. Rats were infused at 3.0 mL/h for 8 h via a duodenal catheter with a lipid emulsion containing 33.3 kBq CH labeled with ©ö©ùC-CH, 20.0 ¥ìmol of CH, 396.0 ¥ìmol of Na-taurocholate, 451.8 ¥ìmol of triolein (oleic acid), 3.1 ¥ìmol alpha-tocopherol and 75.4 ¥ìmol retinol without (control group) or with 240.0 mg AE (AE group) in phosphate-buffered saline (PBS) buffer (pH 6.4). Lymph was collected hourly for 8 h. No significant difference was noted in lymph flow. However, the lymphatic absorption of ©ö©ùC-CH for 8 h was significantly lower in rats infused with AE (34.53 ¡¾ 3.03%) than in control (39.20 ¡¾ 2.87%). Oleic acid in fatty acid output for 8 h in control and AE group was 864.36 ¡¾ 65.27 ¥ìmol and 820.12 ¡¾ 72.88 ¥ìmol, respectively. Total fatty acids were 1,071.43 ¡¾ 68.28 ¥ìmol (control) and 1,015.64 ¡¾ 78.85 ¥ìmol (AE group). Oleic acid and total fatty acids were not significantly decreased by AE infusion but implied reduced intestinal absorption. These data indicate that AE has significant inhibitory effects on the intestinal absorption of CH. Additional studies of intestinal absorption of lipid under different conditions are needed.
