The rumen microbiota of reindeer (Rangifer tarandus) with clinical manifestations of necrobacteriosis | Микробиота рубца у северных оленей (Rangifer tarandus) с клиническими проявлениями некробактериозов

Inglés. Necrobacteriosis is an infectious disease that affects many species of domestic and wild mammals, birds, and humans. The purpose of the study was to characterize the distinctive features of rumen microbiota in the reindeer with the clinical manifestations of necrobacteriosis. The study was carried out on calves (4-6 months) and adults (3-6 years) animals, including clinically healthy individuals and those with necrobacteriosis. Samples of the rumen content were collected during the summer-autumn period in 2017 (n = 3 from each age group) in the Yamalo-Nenets Autonomous District. The total number of bacteria and fungi of the class Neocallimastigales was analyzed by quantitative PCR, the composition of the bacterial community was analyzed by T-RFLP method. In individuals with clinical manifestations of necrobacteriosis, a significantly higher content of fusobacteria was detected, 1.79-fold in adults, and 2.65-fold in calves. In sick animals of both age groups, there was a significantly higher presence of bacteria of the genus Staphylococus and the family Pseudomonadaceae, some species of which may cause purulent-necrotic lesions of animals. The 4-6 month old calves showed a significant increase in the content of family Campylobacteriaceae and family Enterobacteriaceae compared to clinically healthy animals. At the same time healthy individuals showed a greater number of cellulolytic and acid-utilizing bacteria. In general, the rumen microbiome of calves with clinical signs of necrobacteriosis is characterized by large changes compared to the adult animals. In addition, a significant decrease of cellulolytic chytridiomycetes of the class Neocallimastigales was detected in the rumen of this animal group. In this regard, the identified patterns may be due to the physiological features of this stage of animal development in Rangifer tarandus.

Ruso. Некробактериоз - инфекционное заболевание, поражающее многие виды домашних и диких млекопитающих, птиц, человека. Цель исследования - характеристика микробиоты рубца северного оленя при некробактериозе с использованием молекулярных методов. Объектом исследования были северные олени ненецкой породы (здоровые и с признаками некробактериоза), в том числе телята (4-6 мес) и взрослые животные (3-6 лет). Образцы содержимого рубца отбирали в летне-осенний период в 2017 г. (n = 3 для каждой возрастной группы) в Ямало-Ненецком автономном округе. Общее число бактерий и грибов класса Neocallimastigales анализировали методом количественной ПЦР, состав бактериального сообщества - методом Т-RFLP. У особей с клиническими проявлениями некробактериоза детектировано достоверно более высокое содержание фузобактерий: у взрослых оленей в 1,79 раза, у телят - в 2,65 раза. У больных животных обеих возрастных групп отмечали более значительное присутствие бактерий р. Staphylocoссus и сем. Pseudomonadaceae, часть видов которых могут становиться причиной гнойно-некротических поражений. У 4-6-месячных телят выявлено достоверное увеличение численности кампилобактерий (сем. Сampylobacteriaceae) и энтеробактерий (сем. Enterobacteriaceae) по сравнению с клинически здоровыми животными. При этом у здоровых особей обнаружено больше целлюлозолитических бактерий класса Clostridia (в т. ч. представителей сем. Lachnospiraceae, Eubacteriaceae и Clostridiaceae), филума Bacteroidetes и бактерий порядка Negativicutes, ферментирующих кислоты. В целом микробиом рубца телят с клиническими признаками некробактериоза характеризовался большими изменениями по сравнению со взрослыми животными. Также в рубце животных из этой группы было выявлено снижение численности целлюлозолитических грибов-хитридиомицетов класса Neocallimastigales. Выявленные закономерности могут быть обусловлены физиологическими особенностями данной стадии развития северного оленя.
