Immunological detection of sandal spike phytoplasma

Spike disease, caused by a non-culturable phytoplasma is the most serious disease of sandal,Santalum album L., known for its scented heartwood. An immunological method was developed for early detection of the pathogen in infected plants. The phytoplasma was purified by a differential filtration technique and ultra-centrifugation. The purity of the phytoplasma pellet was confirmed using scanning and transmission electron microscopy. Polyclonal antibodies raised in rabbits against purified phytoplasma were used to detect the pathogen present in sandal using direct ELISA technique. The effect of duration on antigen coating and incubation with conjugated antibody in ELISA plates showed that three hours duration was optimum.
