DALP (Direct Amplification Of Length Polymorphism): a PCR based DNA fingerprinting method in brown planthopper, Nilaparvata lugens (STAL) (Homoptera : Delphacidae)

DALP is a new DNA fingerprinting method, based on the use of arbitrarily primed PCR (AP-PCR) to selectively amplify discrete fragments of DNA from the genome of any species which are ready to be sequenced to provide information on different polymorphic loci without having to do cloning. This method was found to be useful in brown planthopper, N. lugens. An oligonucleotide pair DALP235 and DALPR'5 was tested on two sympatric populations of brown planthopper, N. lugens, one from rice (Oryza sativa) and the other from a weed grass (Leersia hexandra) to study the genetic variation between the two populations. This oligonucleotide pair produced polymorphic codominant bands. These codominant bands may be used for the development of microsatellite primers for the brown planthopper, N. lugens.
