Influence of glucoamylase dosage and hydrolysis time on the dextrose equivalent of highly concentrated glucose syrups of extruded starch | Влияние факторов дозировки глюкоамилазы и времени гидролиза на декстрозный эквивалент высококонцентрированных глюкозных сиропов экструдированного крахмала

An alternative to the traditional technology of sugar products from starch based on multistage processes of water-thermal and enzymatic treatment is the use of thermoplastic extrusion with an biocatalysis system integrated in it. As a part of the technology development, a study of the influence of hydrolysis factors on the dextrose equivalent of the obtained sugar products of biocatalysis and their rheological properties was carried out. An important aspect of the research is the study of the process of hydrolysis at high substrate concentration of the medium – 50% of dry matter. The principle of processing highly concentrated media in biotechnology has technical and economic prerequisites, promotes reducing the consumption of heat and energy resources and increases the efficiency of using vessel equipment. Mathematical models of the dependence of the dextrose equivalent and the dynamic viscosity of hydrolysates on the dosage of the saccharifying enzyme glucoamylase and the processing time were derived on the basis of the experiments carried out during the study. The analysis of the derived models showed the equivalence of the influence of both factors on the studied output parameters. The maximum value of the dextrose equivalent in the experiment was 95, corresponding to a glucoamylase dosage of 12 units of glucoamylase per 1 g of starch and 8 h of incubation. Varying the glucoamylase dosage from 6 to 12 units and hydrolysis time from 4 to 8 h allows obtaining products with dextrose equivalent in the range from 38 to 95. A significant decrease in the dynamic viscosity of hydrolysates after 4 h of hydrolysis up to 0.18–0.26 Pa*s relative to the initial values of 2.5–2.6 Pa*s was noted. The obtained models of changes in the degree of hydrolysis and rheology of hydrolysates can be used for estimating and varying the input factors when preparing syrups from extruded starch with a given value of the dextrose equivalent.

Ruso. Альтернативой традиционной технологии сахаристых продуктов из крахмала, основанной на многостадийных процессах водно-тепловой и ферментативной обработки, является использование термпопластической экструзии с интегрированной в нее системой биокатализа. В рамках развития технологии проведено исследование влияния факторов гидролиза на декстрозный эквивалент получаемых сахаристых продуктов и их реологические свойства. Важным аспектом исследования является изучение процесса гидролиза в условиях высокой концентрации среды – 50% сухих веществ. Принцип переработки высококонцентрированных сред в биотехнологии имеет технико-экономические предпосылки, способствует снижению потребления тепло- и энергоресурсов, повышает эффективность использования емкостного оборудования. На основании проведенных в исследовании экспериментальных работ получены математические модели зависимости декстрозного эквивалента и динамической вязкости гидролизатов от дозировки осахаривающего фермента глюкоамилазы и времени обработки. Анализ полученных моделей показал равнозначность влияния обоих факторов на изучаемые выходные параметры. Максимальное значение декстрозного эквивалента в опыте составило 95, соответствующее дозировке глюкоамилазы 12 ед. ГлС/г крахмала и 8 ч инкубирования. Варьирование дозировки глюкоамилазы от 6 до 12 ед. ГлС/г крахмала и времени гидролиза от 4 до 8 ч позволяет получать продукты с декстрозным эквивалентом в диапазоне от 38 до 95. Отмечено значимое снижение динамической вязкости гидролизатов после 4 ч гидролиза до 0,18–0,26 Па*с относительно начальных значений 2,5–2,6 Па*с. Полученные модели изменения степени гидролиза и реологии гидролизатов могут использоваться для оценки и варьирования входных факторов при получения сиропов из экструдированного крахмала с заданным значением декстрозного эквивалента.
