Effect of the silybum marianum extract in experimental diabetic rats on the sera 8-hydroxy-2'-deoxyguanosine, total oxidant and antioxidant capacity

The aim of this study was to find out the effect of Silybum marianum L extract (S marianum) on the 8-hydroxy-2'-deoxyguanosine (8-OHdG), total oxidant (TOS), and antioxidant capacity (TAS) activity. In addition, some other biochemical parameters analyzing such as glucose HbA1c lipoprotein, ALT, AST were performed. In this purpose (28) experimental male rats were prepared in the animal house divided into four groups: first group given normal standard diet and determined as control group (C), second group is diabetic (D) non S. marianum extract treated, third group healthy with S. marianum treated (S) and forth group diabetic with S. marianum treated (DS) Diabetes induced by injecting (45 mg/kg) Streptozotocin intraperitoneally in the D group and DS group. The rats in the S group, and DS. group were treated for 14 days with S marianum L extract dissolved in olive oil 200 mg/kg/day, C group in addition to stander diet oil olive were given. Intragastric gavage was used after this period the parameters such as, HbA1c in whole blood, and glucose HDL, LDL, VLDL, cholesterol, triglyceride, ALT, AST TOS, TAS, 8-OHdG in the serum were measured. Glucose levels and (%HbA1c) were significantly increased (p<0.01) in the D group, decreased significantly. (p<0.01) in DS group. The levels of total cholesterol, triglyceride, LDL, VLDL were significantly increased (p<0.01) in the D group, decreased significantly (p<0.01) in DS group. ALT, and AST levels increased significantly in the D group, decreased significantly (p<0.01) in the DS group. TOS levels increased significantly in the D group (p<0.01), decreased significantly (p<0.01) in DS group. TAS level decreased significantly in the D group (p<0.01), increased significantly (p<0.01) in DS group. The 8-OHdG activity increased significantly (p<0.05) in the D group, decreased but not significantly (p>0.05) in DS group. In conclusion, in this study S. marianum L extract demonstrated remarkable anti-diabetic activity, also demonstrated its ability to decrease diabetic dyslipidemia. This plant protecting cells against oxidative stress, increasing the non-enzymatic and enzymatic antioxidant defense systems of cells. Finally, S. marianum L extract affected on 8-OHdG but not significant.
