The objective of this research was to study the efficiency of certain plant growth regulators on in vitro shoot, root and callus induction in the medicinal plants: Phaya Yo \"Dong Bang\" (Clinacanthus nutans (Burm.f.) Lindau.). The node explants were surface sterilized with 10 percent and 5 percent chlorox solution, respectively. Then the explants were cultured on Murashige and Skoog (MS) medium supplemented with growth regulators: Thidiazuron (TDZ) at the concentrations of 0, 0.5 and 1.0 mg/L in combination with 1-Naphthaleneacetic acid (NAA) at the concentrations of 0, 0.01 and 0.1 mg/L for 2 months. The results showed that the media with NAA 0, 0.01 and 0.1 mg/L showed elongated green shoots from nodes, with the average shoot number of 1-1.4 plants/node and average height of 4.22-4.79 cm. While MS with 0.5-1.0 mg/L TDZ or in combination with 0, 0.01 and 0.1 mg/L NAA resulted of multiple shoots production at the average number of 4.0-6.33 plants/node. The optimum multiple shoots induction medium was MS medium with 1.0 mg/L TDZ and 0.1 mg/l NAA. Multiple shoots derived from the media with 0.5 or 1.0 mg/L TDZ alone or in combination with 0, 0.01 and 0.1 mg/L NAA were subcultured to the same media and cultured for 2 months. It was found that some leaves fell down and compact yellowish-green calli were occurred and expanded at the base of multiple shoot in all media. After that yellowish-green calli developed to numerous green shoot primordia. It was observed that the medium with 1.0 mg/L TDZ alone was the most effective in stimulating mass calli formation and developing a large number of shoots primordia. The newly shoots from the media containing 0, 0.01 and 0.1 mg/L NAA could induce root initiation. The best root induction medium was MS medium with 0.1 mg/L NAA. The new plantlets were transplanted into the pot containing soil mixture and kept in the nursery. It showed 100 percent of survival rate. The gained technology could be used to optimize the production of multiple shoots and to stimulate mass production of calli with shoots primordia initiation in the laboratory. This project will be the basis for further research on the production of useful medically bioactive compounds from tissues or calli of Phaya Yo \"Dong Bang\" herb in sterile condition
