Regeneration and multiplication of black diamond and myrobolan 29 C plums

Two important fruit species among Prunus plants, European Plums (Prunus domestica) and Japanese Plums (Prunus salicina) are gradually becoming important plants worldwide. In this thesis, both Black Diamond plum species which belong to Japanese group and Myrobolan 29 C plum rootstocks which are a part of European group are studied for regeneration using shoots and leafs, while a multiplication study is carried out only for Myrobolan 29 C.In the multiplication studies for Myrobolan 29 C in WPM medium, the effects of different doses of BA and TDZ growth regulators on shoot multiplication are observed. The best results of multiplication with different concentrations of BA were obtained from 1,25μM BA (% 94,2) and 2,5 μM BA (% 94,2), followed by 5,0 μM BA (% 85,7). Multiplication with different doses of TDZ gave its best result from the medium containing 1 mgl-1 TDZ with the ratio of % 85.In the regeneration experiments, the effects of plant growth regulators, light regimes and pretreatments on leaf regeneration are investigated. Regeneration couldn't be achieved from Black Diamond with BA and TDZ applied in both dark and light environments. However, applications of TDZ and BA to Myrobalan 29C rootstock, regeneration is observed from mediums containing 2,5 μM BA (% 2,5), 1 mgl-1 TDZ (% 2,5) and 2 mgl-1 TDZ (% 2,5) in light environment. In dark environment, regeneration is observed from mediums containing 2,5 μM BA (% 2,5), 5 μM BA (% 2,5), 0,5 mgl-1 TDZ and 0.25 mgl-1 TDZ. The plants obtained from regeneration are left rooting in an 2 mgl-1 IBA medium, All rooted plantes were acclimatized and then transferred to the pots.
