In vitro regeneration and multiplication of Maxma14 (Prunus mahaleb L. × P. avium L.) and Mazzard F12-1 (P. avium L.) cherry rootstocks

In this thesis, in vitro multiplication and regeneration of Mazzard F 12-1 and Maxma 14 rootstocks were studied. In multiplication studies, effect of BA, TDZ and their different concentration on in vitro shoot propagation were investigated. MS medium was used in these studies. The best results on multiplication obtained from 20 mg l?¹ BA (1.40 /explant), 10 mg l?¹ BA(1,20 /explant), 5 mg l?¹ BA (1,36 /explant), 2,5 mg l?¹ BA (1,08 /explant), 0,125 mg l?¹ TDZ (1,03 /explant), 0,25 mg l?¹ TDZ (1,52 /explant) and 0,5 mg l?¹ TDZ (1,16 /explant). In Maxma 14, the best result were obtained from 2 mg l?¹ TDZ (1,2 /explant), 1,5 mg l?¹ TDZ (1,03 /explant), 1 mg l?¹ TDZ (0,96 /explant), 20 mg l?¹ BA (1,26 /explant), and 10 mg l?¹ BA (1,10 /explant).Regeneration studies carried out on WPM medium containing 0,54 µM NAA. In regeneration studies, the effects of Plant Growth Regulators (PGR), dark and pre-treatments were investigated. No shot regeneration was obtained in Maxma 14. In Mazzard F 12-1 %5 shoot regeneration was obtained in BA 5 mg l?¹, BA 10 mg l?¹ and TDZ 2 mg l?. Effect of dark incubation (5, 10, 15 and 20 days) on regeneration was also investigated and % 2,5 regeneration was obtained in 5 and 15 days dark incubation. Optimization studies with TDZ and BA were carried out to increase the regeneration rates without success. IBA was used to the rootstocks and the best rooting percentages were obtained with 2 mg l?¹ IBA and 1 mg l?¹ IBA. Shoots obtained from regeneration studies were rooted in MS medium containing 2 mg l?¹ IBA then transferred to soil and acclimatized.
