Identification of ´candidatus phytoplasma mali´ genotypes using imp gene and their geographical distribution in the Czech Republic

The work summarizes the results of the study of genetic variability of ´Candidatus Phytoplasma mali´in the Czech Republic using imp gene that codes immunodominant membrane protein. Eightyone imp gene sequences were obtained out of 98 phytoplasma positive samples. Based on the sequence alignment, seven distinct imp genotypes were defined belonging to two groups: A1 to A3, and B1 to B4. Interestingly, three imp genotypes (A2, A3, and B2) have been described for the first time. In 15 samples (18.5 %), mixed infections of various imp genotypes were noticed. Sequence similarity between ´Candidatus Phytoplasma mali´ imp genotypes was 90.5 % to 99.8 %. Based on symptoms assessment, it is impossible to discriminate infected trees from uninfected with certainty, since 63.6 % phytoplasma negative samples showed symptoms, while 41.1 % phytoplasma positive samples were asymptomatic. A correlation between ´Candidatus Phytoplasma mali´ imp genotype and symptoms was not observed.
