The study of nuclear techniques for rice root studies by using phosphorus-32

Four methods using P-32 namely (1) injecting P-32 solution; (2) dropping ice cube of P-32; (3) labelling 3 gm-soil with P-32 and (4) injecting P-32 solution into mud ball, for studying root systems and root activity of rice plant were studied. All methods were tested under saturated moist soil in thin wooden boxes having one side attached with plastic sheet. The X-ray film (Kodak no-screen film ready pack NS-2T) was immediately placed in the plastic sheet where P-32 spot was, followed by flooding water into the boxes and were kept for 7 days. The labelled spot was confirmed by autoradiography with 7 days exposure time. The area of the black spot which appeared on the X-ray film was calculated. The techniques were tested at three different times. All methods gave the same results and are possible to use as techniques in studying root system and root activity of rice plant. However, the most practical method is the injection P-32 into the soil.
