Screening of fungal strains for protease production

Based on measured kinetic properties of the proteases Aspergillus niger protease was found to be the most suitable strain. Therefore A. niger was selected as the most productive strain based on its protease activity in the solid synthetic medium, liquid synthetic medium and date extract medium. Protease activity levels of this strain were similar to those of A. niger ATCC 16888 (reference strain) at various screening stages. A. niger protease has an optimum pH of 4.0 and was most stable at pH range of 4.0-6.0. The enzyme retained 66% of its original activity when incubated for 30 min at 60%
