Genomic variation among field isolates of Rhizobium trifolii

A series of samples of rhizobia were isolated from the nodules of white clover collected from a small long-established plot at the Massey Tuapeka Unit, New Zealand, and subjected to the standard procedures of genomic DNA analysis. It was shown that a geographically restricted population of Rhizobium can manifest substantial, though not infinite, variability at the level of plasmid profile. There is also substantial variation in genomic restriction digest profiles. The variability of plasmid profile is not necessarily accompanied by variability at the level of genomic restriction digest profile. Radiolabelled nifKDH probes of both plasmid and genomic restriction digest profiles show a high degree of conservation on the sampled population, of both the plasmid location and the nucleotide sequence neighbourhood of these nitrogen fixation genes
