Some ecological aspects of the relationship between grapevine fanleaf virus and its nematode vector Xiphinema index [Portugal]

Ecological studies on the relationship between the nematode vector X. index and Grapevine Fanleaf Virus were carried out in a vineyard by using ELISA in the diagnosis of the virus. Vertical distribution of nematodes and GFV antigen in roots was studied. High numbers of infected nematodes were found at deeper layers of the soil (90-120 cm), associated to infected roots. ELISA technique was adequate to disclose GFV in X. index and roots, 25 nematodes being sufficient to give reliable absorbance values. A defined pattern of distribution of the A405 values in roots was not found in the soil profile. ELISA values were probably related to the lignification degree of the roots. The presence of the GFV in roots and nematodes at deeper layers of soil makes the control measures particularly difficult (author)
