Tuberisation "in vitro" of potatoes

Tuberisation in vitro was studied with the local cultivars (Vesna, Igor, Cvetnik, Jana) and foreign cv. Desiree. The influence of phytohormones and length of photoperiod on microtuber induction was studied. Microplants were grown on control medium A - modified MS (1962) media containing 8 % sucrose and on medium B - supplemented with growth regulators (BAP, NAA, CCC), medium WH (BAP) and medium D (KIN). The liquid basic culture medium C was also tested. The growth took place in darkness, under 8 and 16 hrs daylight. The largest microtubers grew on liquid medium. Microtubers of foreign cv. Desiree and local cv. Vesna and Jana were the largest. The influence of genotype of cultivar on the number and weight of microtubers was important. The use of liquid MS medium with 8 % of sucrose (C) in dark or 8 hrs of photoperiod could be recommended for all tested cv. strict arrangement and field crop rotation
