Isolation and confirmation of the presence of corysamine 5,6-dihydrosanguinarine and 5,6-dihydrochelerythrine in roots of Chelidonium majus L. growing in Poland

So far 28 alkaloids have been isolated from Chelidonium majus L., among them corysamine, 5,6-dihydrosanguinarine and 5,6-dihydrochelerythrine. It the literature on the isolation, separation and identification of alkaloids from C. majus growing in Poland there is no information on these alkaloids. Carrying out systematic investigations on isolation and separation of C. majus alkaloids, from the fraction of corysamine was isolated. Identification of 5,6-dihydrosanguinarine and 5,6-dihydrochelerythrine, which were present in fraction of tertiary alkaloids, was carried out by exploitation of the ability of these alkaloids to oxidation in air to corresponding quaternary alkaloids, sanguinarine and chelerythrine. Spontaneous oxidation of these alkaloids can be easily performed by leaving their spots on the silica gel layer for 2-3 hours, which resulted in formation of a coloured spot of sanguinarine and chelerythrine. Additionally the identity of dihydrosanguinarine and dihydrosanguinarine was proved by comparoson of Rf values, fluorescence in UV light at 366 nm and two-dimensional TLC before oxidation and after oxidation of the dihydroderivatives
